SP1 antibody targets Specificity Protein 1 (SP1), a transcription factor belonging to the SP/KLF family. Discovered in the 1980s, SP1 binds GC-rich promoter regions (GC-boxes) via three zinc finger domains, regulating genes involved in cell growth, differentiation, and apoptosis. It interacts with coactivators/corepressors (e.g., p300. HDACs) and modulates chromatin structure. SP1 is ubiquitously expressed but dysregulated in cancers (e.g., overexpression in tumors), neurodegenerative diseases, and viral infections (e.g., HPV, HIV). Its role in oncogenesis includes promoting cell proliferation (via cyclin D1. VEGF) and inhibiting apoptosis (via survivin). SP1 antibodies are essential tools in research—used in techniques like ChIP, Western blot, and immunofluorescence to study SP1-DNA/Protein interactions, expression patterns, and subcellular localization. Commercial SP1 antibodies (polyclonal/monoclonal) vary in specificity; validation via knockout controls is recommended. Dysregulation studies using these antibodies have informed therapeutic strategies targeting SP1 activity in cancer and inflammation.
