背景信息

Glycophorin C (GPC) is a transmembrane glycoprotein predominantly expressed on the surface of human erythrocytes. It plays a structural role in maintaining erythrocyte membrane integrity by interacting with the cytoskeleton via protein 4.1R. GPC is encoded by the *GYPC* gene and serves as a receptor for the *Plasmodium falciparum* malaria parasite in some individuals. It also carries blood group antigens, notably the Gerbich (Ge) system, which includes Ge2. Ge3. and Ge4 epitopes. Antibodies targeting Glycophorin C are clinically significant in transfusion medicine and hematology. Anti-GPC antibodies may arise naturally or through alloimmunization (e.g., after blood transfusions or pregnancy). These antibodies are associated with hemolytic transfusion reactions or neonatal alloimmune neutropenia in rare cases. In laboratory diagnostics, anti-GPC antibodies are utilized to identify erythrocyte membrane defects, such as hereditary elliptocytosis or Southeast Asian ovalocytosis, where GPC expression may be altered. Monoclonal antibodies against specific GPC epitopes are also employed in flow cytometry to study erythrocyte maturation or membrane disorders. Research applications include investigating malaria pathogenesis, as GPC-mediated invasion mechanisms offer potential therapeutic targets. Commercial GPC antibodies are typically raised in mice or rabbits using purified protein or peptide antigens, validated for techniques like Western blotting, immunofluorescence, or agglutination assays.
