LAMP1 (Lysosome-Associated Membrane Protein 1) is a key transmembrane glycoprotein predominantly localized in the membranes of lysosomes and endosomes. It plays a critical role in maintaining lysosomal integrity, regulating vesicular trafficking, and participating in autophagy and phagocytosis. Structurally, LAMP1 contains a heavily glycosylated luminal domain, which protects lysosomal membranes from enzymatic degradation, and a short cytoplasmic tail that mediates interactions with cytoskeletal components and signaling molecules.
Antibodies targeting LAMP1 are widely used as markers to identify lysosomal compartments in immunofluorescence, immunohistochemistry, and flow cytometry. In research, they help study lysosomal dysfunction linked to neurodegenerative diseases (e.g., Alzheimer’s, Parkinson’s), cancer (lysosomal exocytosis in metastasis), and infectious diseases (pathogen-lysosome interactions). LAMP1 is also referred to as CD107a, with its surface expression serving as an indicator of immune cell degranulation (e.g., cytotoxic T cells, NK cells). Additionally, LAMP1 antibodies aid in exploring cellular responses to stressors like pH changes or nutrient deprivation, providing insights into lysosomal adaptation mechanisms. Their utility spans basic cell biology, drug development, and diagnostic research, underscoring LAMP1's significance in cellular homeostasis and disease pathology.
