Prohibitin (PHB) is a highly conserved, ubiquitously expressed protein belonging to the stomatin/prohibitin/flotillin/HflK/C (SPFH) domain family. Initially identified as an antiproliferative protein in mammalian cells, PHB is localized primarily in the mitochondrial inner membrane, where it plays critical roles in mitochondrial homeostasis, including maintaining structural integrity, regulating oxidative phosphorylation, and chaperoning protein folding. It also shuttles to the nucleus, cytoplasm, or cell membrane under specific conditions, participating in diverse cellular processes such as apoptosis, senescence, and transcriptional regulation.

Prohibitin antibodies are essential tools for studying its dual roles in health and disease. Two isoforms, PHB1 (~32 kDa) and PHB2 (~37 kDa), form a heterodimeric complex in mitochondria.
