**Background of ZAP70 Antibodies**
ZAP70 (zeta-chain-associated protein kinase 70) is a cytoplasmic tyrosine kinase belonging to the Syk family, primarily involved in T-cell receptor (TCR) signaling. It plays a critical role in immune responses by mediating signal transduction after TCR activation. Structurally, ZAP70 contains two N-terminal SH2 domains that bind phosphorylated immunoreceptor tyrosine-based activation motifs (ITAMs) on the CD3ζ chain of the TCR complex, and a C-terminal kinase domain responsible for downstream signaling.
ZAP70 antibodies are essential tools in research and diagnostics. In research, they help study T-cell development, activation, and dysfunction, particularly in immunodeficiencies or autoimmune diseases. Clinically, ZAP70 expression is a prognostic marker in chronic lymphocytic leukemia (CLL), where its presence correlates with aggressive disease and unmutated immunoglobulin heavy-chain genes. Antibodies against ZAP70 are used in flow cytometry, Western blotting, and immunohistochemistry to detect protein expression levels and cellular localization.
Mutations in the *ZAP70* gene are linked to severe combined immunodeficiency (SCID), characterized by dysfunctional T cells. ZAP70 antibodies aid in diagnosing such genetic disorders. Additionally, they contribute to understanding ZAP70's role in other immune cells, like natural killer cells, and its interplay with signaling pathways (e.g., MAPK, PI3K).
Overall, ZAP70 antibodies bridge basic immunology and clinical applications, advancing insights into immune regulation, disease mechanisms, and therapeutic targeting.
