背景信息

The T-box protein 3 (TBX3) is a member of the T-box family of transcription factors, characterized by a conserved DNA-binding domain called the T-box. TBX3 plays critical roles in embryonic development, particularly in regulating organogenesis, limb formation, and cell fate determination. It is known to repress transcription of target genes, including the tumor suppressor p14ARF (in mice) or p19ARF (in humans), thereby influencing cell proliferation, differentiation, and apoptosis. Dysregulation of TBX3 has been implicated in developmental disorders and cancers. For example, mutations in TBX3 are linked to ulnar-mammary syndrome, a congenital condition affecting limb, apocrine gland, and genital development. In cancer, TBX3 overexpression is associated with tumor progression, metastasis, and drug resistance in melanoma, breast, and liver cancers.

Antibodies targeting TBX3 are essential tools for studying its expression, localization, and function in both normal and pathological contexts. These antibodies are typically generated using immunogens derived from specific regions of the human or murine TBX3 protein, such as the N-terminal or T-box domain. They are widely used in techniques like Western blotting, immunohistochemistry (IHC), immunofluorescence (IF), and chromatin immunoprecipitation (ChIP) to investigate TBX3's role in gene regulation, developmental pathways, and oncogenic mechanisms. Validation of TBX3 antibodies includes testing for specificity via knockout cell lines or siRNA-mediated knockdown. Researchers also utilize these antibodies to explore TBX3's potential as a diagnostic marker or therapeutic target, particularly in cancers where its aberrant expression correlates with poor prognosis.
