背景信息

Rel B, a member of the NF-κB transcription factor family, plays a distinct role in immune regulation and cellular responses. Structurally, it contains a Rel homology domain (RHD) for DNA binding and dimerization, and a transactivation domain (TAD) for gene activation. Unlike canonical NF-κB proteins like RelA, Rel B primarily operates in the non-canonical pathway, forming complexes with p100 (processed to p52) to regulate gene expression. This pathway is activated by specific receptors, such as CD40. BAFF-R, or lymphotoxin-β receptor, leading to slow but sustained signaling.

Rel B is critical for secondary lymphoid organ development, immune cell differentiation, and adaptive immunity. It promotes dendritic cell maturation, B-cell survival, and T-cell homeostasis. Additionally, it modulates genes involved in inflammation, apoptosis, and immune tolerance. Its activity is tightly regulated through interactions with inhibitory proteins (e.g., p100) and post-translational modifications.

In disease contexts, Rel B exhibits dual roles. It contributes to autoimmune disorders (e.g., rheumatoid arthritis) by sustaining inflammation, while in cancer, it may act as an oncogene or tumor suppressor depending on the context—for example, promoting survival in prostate cancer or suppressing metastasis in certain lymphomas. Rel B-specific antibodies are essential tools in research, enabling detection of its expression, localization, and activation via techniques like Western blotting, immunohistochemistry, and flow cytometry. Studying Rel B provides insights into immune dysregulation and therapeutic targeting for inflammatory diseases and malignancies.
