背景信息

ORP150 (Oxygen-Regulated Protein 150), also known as GRP170 or HYOU1. is a stress-inducible molecular chaperone belonging to the heat shock protein 70 (HSP70) family. It was first identified in the 1990s as a protein upregulated under hypoxic conditions, playing a critical role in cellular adaptation to low oxygen tension and endoplasmic reticulum (ER) stress. Structurally, ORP150 contains an N-terminal ATPase domain and a C-terminal substrate-binding domain, enabling its participation in protein folding, quality control, and the unfolded protein response (UPR).

This 150 kDa protein is predominantly localized in the ER and is essential for maintaining cellular homeostasis during stress by stabilizing nascent proteins, preventing aggregation, and aiding in the degradation of misfolded proteins. Its expression is regulated by hypoxia-inducible factor 1 (HIF-1α) and other stress-sensitive signaling pathways.
