背景信息

Nucleophosmin (NPM), also known as B23 or NPM1. is a multifunctional phosphoprotein primarily localized in the nucleolus. It plays critical roles in ribosome biogenesis, centrosome duplication, cell cycle regulation, and genomic stability. NPM shuttles between the nucleolus, nucleus, and cytoplasm, participating in processes like chromatin remodeling, DNA repair, and apoptosis. Its involvement in diverse cellular pathways makes it a key focus in cancer research, particularly in hematologic malignancies.

NPM antibodies are essential tools for detecting and studying this protein in research and diagnostics. In normal cells, NPM antibodies help visualize nucleolar organization and monitor protein dynamics during stress or cell division. However, their clinical significance is most notable in acute myeloid leukemia (AML), where mutations in the NPM1 gene (exon 12 insertions) result in aberrant cytoplasmic localization of NPM (NPMc+). Antibodies specific to mutant NPM are used diagnostically to identify this AML subtype, which correlates with prognosis and treatment response.

Beyond cancer, NPM antibodies aid in studying viral infections, neurodegenerative diseases, and ribosomopathies. They are employed in techniques like immunohistochemistry, Western blotting, and immunofluorescence. Challenges include distinguishing between NPM isoforms and ensuring specificity due to post-translational modifications. Ongoing research aims to refine antibody-based assays for precision medicine, underscoring NPM's role as both a biomarker and a therapeutic target.
