Lactate dehydrogenase C (LDH-C), also known as LDHx or LDH3. is a tissue-specific isoform of the lactate dehydrogenase enzyme family, primarily expressed in germ cells of the testes and mature spermatozoa. Unlike other LDH isoforms involved in glycolysis, LDH-C plays a specialized role in sperm energy metabolism and maturation. Its unique structure, encoded by the LDHC gene on chromosome 11. allows optimal function in the anaerobic environments of the male reproductive tract.
Anti-LDH-C antibodies are autoantibodies that target this isoform. They may arise due to immune dysregulation, often linked to conditions like autoimmune infertility or testicular injury. In males, these antibodies can impair sperm function by interfering with energy production, potentially leading to reduced motility or immunoinfertility. In females, anti-LDH-C antibodies have been associated with immune responses against sperm antigens, contributing to unexplained infertility.
Clinically, LDH-C antibodies are investigated as biomarkers for immune-related infertility. Research also explores their relevance in certain cancers, as some tumors aberrantly express LDH-C to support Warburg-effect metabolism. Emerging studies suggest diagnostic potential in testicular cancer and chemotherapy resistance monitoring. Additionally, LDH-C's immunogenicity has prompted investigations into contraceptive vaccines, though therapeutic applications remain experimental. Current detection methods include ELISA and immunoblotting, primarily in research settings.
