Aberrant expression of AZI2 in dendritic cells promotes carcinogenesis by suppressing antitumor immunity"*

Y. Matsumoto et al.

The AZI2 (5-azacytidine-induced protein 2), also known as CARD-containing MAGUK protein 3 (CARMA3), is a scaffold protein involved in immune signaling pathways. Structurally, it belongs to the membrane-associated guanylate kinase (MAGUK) family, characterized by a PDZ domain, an SH3 domain, and a catalytically inactive guanylate kinase (GK) domain. Additionally, AZI2 contains a caspase recruitment domain (CARD), enabling interactions with downstream signaling molecules. It plays a critical role in regulating NF-κB activation, particularly in response to T-cell receptor (TCR) engagement or Toll-like receptor (TLR) stimulation. AZI2 facilitates the assembly of signaling complexes by bridging adaptor proteins like BCL10 and MALT1. which are essential for inflammatory and immune responses. Dysregulation of AZI2 has been implicated in autoimmune disorders, lymphomas, and inflammatory diseases. Antibodies targeting AZI2 are valuable tools for studying its expression, localization, and function in immune cells. They are widely used in techniques such as Western blotting, immunofluorescence, and co-immunoprecipitation to dissect its role in signal transduction and pathological contexts. Research on AZI2 continues to shed light on its therapeutic potential in modulating immune-related diseases.
