Integrin alpha IIb (αIIb), also known as CD41. is a key subunit of the heterodimeric transmembrane receptor integrin αIIbβ3 (GPIIb/IIIa), predominantly expressed on platelets and megakaryocytes. As a member of the integrin family, it plays a critical role in mediating cell adhesion, signal transduction, and platelet aggregation during hemostasis and thrombosis. The αIIbβ3 complex binds ligands such as fibrinogen, von Willebrand factor, and fibronectin upon platelet activation, facilitating cross-linking of adjacent platelets and clot formation.
Antibodies targeting integrin alpha IIb are essential tools in studying platelet biology and related pathologies. They are widely used in research applications, including flow cytometry, immunofluorescence, and Western blotting, to detect αIIb expression or monitor its activation state. Dysregulation of αIIbβ3 is implicated in bleeding disorders like Glanzmann thrombasthenia (caused by genetic defects in αIIb or β3) and thrombotic diseases. Anti-αIIb antibodies also contribute to diagnosing these conditions and evaluating therapeutic interventions, such as αIIbβ3 antagonists (e.g., abciximab) used to prevent clotting in cardiovascular procedures. Additionally, they aid in investigating megakaryocyte development and platelet production. Their specificity makes them valuable for distinguishing αIIb from other integrin α subunits, enabling precise mechanistic studies in hematopoiesis, thrombosis, and inflammation.
