背景信息

TALL-2. also known as BAFF receptor (BAFF-R) or TNFRSF13C, is a transmembrane protein belonging to the tumor necrosis factor receptor superfamily. It primarily binds to B cell-activating factor (BAFF, also called BLyS), a cytokine critical for B cell survival, maturation, and homeostasis. Discovered in the early 2000s, TALL-2 is predominantly expressed on mature B lymphocytes and plays a key role in the BAFF/BAFF-R signaling axis, which regulates adaptive immunity and immune tolerance. Dysregulation of this pathway is linked to autoimmune diseases, such as systemic lupus erythematosus (SLE) and rheumatoid arthritis, where excessive BAFF signaling promotes autoreactive B cell proliferation and autoantibody production. Structurally, TALL-2 contains cysteine-rich extracellular domains for ligand binding and intracellular regions that recruit TRAF adaptor proteins to activate downstream NF-κB and MAPK pathways. Research on TALL-2 antibodies focuses on therapeutic strategies to block pathogenic BAFF interactions, with some agents (e.g., belimumab, an anti-BAFF monoclonal antibody) already approved for autoimmune conditions. TALL-2-targeting antibodies are also explored as tools to study B cell biology or potential treatments for B cell malignancies. However, challenges remain in balancing therapeutic efficacy with risks of immunosuppression.
