The p14 ARF (Alternative Reading Frame) protein, encoded by the INK4a/ARF locus on human chromosome 9p21. is a critical tumor suppressor involved in cell cycle regulation and apoptosis. This locus uniquely encodes two distinct proteins, p16 INK4a and p14 ARF, through alternative reading frames. While p16 primarily inhibits CDK4/6 to block Rb phosphorylation, p14 ARF stabilizes p53 by binding to MDM2. thereby preventing p53 degradation and enhancing its tumor-suppressive functions. Dysregulation of p14 ARF, through mutations, deletions, or epigenetic silencing, is frequently observed in cancers, contributing to uncontrolled proliferation and evasion of apoptosis.
