背景信息

The 53BP1 (p53-binding protein 1) antibody is a critical tool in studying DNA damage response (DDR) mechanisms. 53BP1 is a multifunctional protein involved in repairing DNA double-strand breaks (DSBs), particularly by promoting non-homologous end joining (NHEJ) over homologous recombination (HR). It accumulates at DSB sites, forming microscopically detectable nuclear foci, and acts as a scaffold to recruit downstream DDR factors. Dysregulation of 53BP1 is linked to genomic instability, cancer progression, and chemoresistance.

The 53BP1 antibody is widely used in research to visualize and quantify DNA damage foci via immunofluorescence (IF) or immunohistochemistry (IHC). It also serves in Western blotting (WB) to assess protein expression levels. This antibody has become a biomarker for evaluating radiation- or chemotherapy-induced DNA damage in preclinical studies. Additionally, 53BP1's role in regulating DNA repair pathway choice has therapeutic implications, particularly in BRCA-deficient cancers. Loss of 53BP1 can restore HR in BRCA1-mutant cells, conferring resistance to PARP inhibitors, making its detection crucial for predicting treatment outcomes.

Commercial 53BP1 antibodies are typically raised against conserved epitopes, validated for specificity across human and mouse models. Their applications span basic DDR studies, cancer biology, and drug development, underscoring their importance in precision oncology and biomarker discovery.
