The RO60 antibody, also known as anti-SSA2 or anti-Ro60. is an autoantibody targeting the RO60 (SSA2) protein, a RNA-binding protein involved in RNA quality control and cellular stress responses. It is classified under antinuclear antibodies (ANAs) and is clinically associated with systemic autoimmune diseases, particularly systemic lupus erythematosus (SLE) and Sjögren’s syndrome (SS). RO60 forms a complex with Y RNAs, facilitating the recognition and degradation of misfolded noncoding RNAs, and may play roles in regulating gene expression and immune signaling.
Clinically, RO60 antibodies are diagnostic markers for SLE and SS, often correlating with specific manifestations. In SLE, they are linked to photosensitivity, cutaneous involvement, and neonatal lupus—a condition where maternal antibodies cross the placenta, potentially causing congenital heart block or skin lesions in infants. In SS, RO60 antibodies are frequently co-detected with anti-La/SSB antibodies and associated with salivary gland dysfunction and extraglandular complications. Their presence may also indicate increased risk of interstitial lung disease in overlap syndromes like scleroderma.
Detection methods include ELISA, immunoblot, and immunofluorescence. While not disease-specific, RO60 antibodies aid in diagnosis, prognosis, and risk stratification. Research continues to explore their pathogenic role, including molecular mimicry or epitope spreading mechanisms, and their interplay with environmental triggers like ultraviolet light. These studies aim to refine therapeutic strategies targeting autoantibody production or downstream immune pathways.
