Filaggrin (FLG) antibodies are essential tools in studying the role of filaggrin, a key protein in epidermal differentiation and skin barrier function. Filaggrin is encoded by the FLG gene and synthesized as a large precursor protein, profilaggrin, which is stored in keratohyalin granules within the stratum granulosum of the epidermis. During terminal differentiation, profilaggrin is cleaved into functional filaggrin monomers that aggregate keratin filaments, facilitating the collapse of corneocytes into a flattened, protective stratum corneum. Subsequently, filaggrin undergoes further degradation into hygroscopic amino acids that contribute to skin hydration and pH regulation.

Mutations in the FLG gene are strongly associated with impaired skin barrier integrity, leading to disorders like ichthyosis vulgaris and atopic dermatitis (eczema).
