背景信息

CD180. also known as LY64 or RP105. is a cell surface glycoprotein belonging to the Toll-like receptor (TLR) family. It was initially identified as a homolog of the Drosophila Toll protein and shares structural features with TLR4. including leucine-rich repeats and a putative ligand-binding domain. Unlike most TLRs, CD180 lacks the intracellular Toll/interleukin-1 receptor (TIR) signaling domain, functioning instead as a co-receptor that modulates immune cell responses. Primarily expressed on B lymphocytes and antigen-presenting cells, CD180 forms a complex with MD-1 (lymphocyte antigen 86), enabling its surface expression and ligand interaction.

CD180 antibodies are critical tools for studying its role in B cell activation, survival, and tolerance. Research shows CD180 participates in pathogen recognition through undefined ligands, potentially influencing TLR4-mediated signaling pathways. Its engagement by antibodies or natural ligands triggers intracellular cascades involving kinases like PI3K and MAPK, affecting cell proliferation and cytokine production. In autoimmune diseases and lymphomas, altered CD180 expression has been observed, linking it to dysregulated immune responses. Antibodies targeting CD180 are used in flow cytometry, immunohistochemistry, and functional assays to explore its therapeutic potential in immune modulation or cancer treatment. Recent studies also investigate its crosstalk with other receptors, such as CD19 and CD20. in B cell malignancies.
