CD244. also known as 2B4 or SLAMF4. is a cell surface receptor belonging to the signaling lymphocytic activation molecule (SLAM) family. It is primarily expressed on natural killer (NK) cells, CD8+ T cells, and certain T-cell subsets. Structurally, CD244 contains two extracellular immunoglobulin-like domains, a transmembrane region, and a cytoplasmic tail with immunoreceptor tyrosine-based inhibitory motifs (ITIMs) and switch motifs (ITSMs). These motifs enable CD244 to transmit bidirectional signals, modulating immune cell activation or inhibition depending on ligand binding and intracellular adaptor interactions. Its ligand, CD48. is a glycosylphosphatidylinositol (GPI)-anchored protein expressed on immune and infected cells.
CD244 plays a dual role in immune regulation. Engagement with CD48 can enhance NK cell cytotoxicity and cytokine production via ITSM-mediated recruitment of SAP family adaptors. Conversely, ITIM-dependent signaling can suppress immune responses by recruiting phosphatases like SHP-1/SHP-2. particularly in chronic infections or malignancies.
