Neural cell adhesion molecule 1 (NCAM1. also known as CD56) is a transmembrane glycoprotein belonging to the immunoglobulin superfamily. It plays critical roles in cell-cell adhesion, neuronal development, synaptic plasticity, and immune regulation. NCAM1 mediates homophilic binding (NCAM-NCAM interactions) and heterophilic interactions with extracellular matrix components, facilitating cell migration, axon guidance, and tissue morphogenesis. Its expression is prominent in the nervous system, but it is also detected in immune cells (e.g., natural killer cells) and certain cancers, including gliomas, melanomas, and multiple myeloma.
NCAM1 antibodies are essential tools for studying its biological functions and pathological implications. They enable the detection of NCAM1 isoforms (e.g., 120. 140. and 180 kDa variants arising from alternative splicing) and post-translational modifications, such as polysialylation (PSA-NCAM), which modulates cell adhesion. In research, these antibodies are used in techniques like Western blotting, immunohistochemistry, and flow cytometry to explore NCAM1's role in neural repair, cancer metastasis, and immune responses. Clinically, NCAM1 antibodies have diagnostic potential in identifying CD56-positive tumors and are being investigated for therapeutic targeting. Aberrant NCAM1 expression or glycosylation is linked to neurological disorders (e.g., schizophrenia, Alzheimer’s) and cancer progression, underscoring its biomedical relevance.
